A floxedDll4 allele was generated in the following fashion, the loxP-flanked neomycin phosphotransferase (NeoR) gene was inserted after exon 3, and a third loxP site was also placed before the translational initiation site in exon 1. After the NeoR gene was depleted in ES cell clones containing the homologous recombination allele of Dll4, one of these clones was injected into blastocysts and chimeric mice were obtained.
